WATCH R641G

R641G is newly appearing on 10 isolates across 1 region(s) (USA), first seen 2026; detection only -- no structural verdict; echinocandins do not coordinate a metal, so the CYP51/heme-iron geometry does not transfer to FKS1. It sits in hot-spot HS1, 2 residue(s) after the S639 anchor — adjacent (≤2 aa) to known marker position 639.

WATCH S639Y

S639Y is newly appearing on 9 isolates across 2 region(s) (China, USA), first seen 2019-01-24; detection only -- no structural verdict; echinocandins do not coordinate a metal, so the CYP51/heme-iron geometry does not transfer to FKS1.

WATCH F635L

F635L is newly appearing on 6 isolates across 1 region(s) (USA), first seen 2023-04; detection only -- no structural verdict; echinocandins do not coordinate a metal, so the CYP51/heme-iron geometry does not transfer to FKS1. It sits in hot-spot HS1, 4 residue(s) before the S639 anchor — a novel substitution at residue 635, where C/Y are known echinocandin-resistance changes.

WATCH F635Y

F635Y is newly appearing on 5 isolates across 2 region(s) (Colombia, USA), first seen 2025-04; detection only -- no structural verdict; echinocandins do not coordinate a metal, so the CYP51/heme-iron geometry does not transfer to FKS1.

WATCH R1354C

R1354C is newly appearing on 3 isolates across 1 region(s) (USA), first seen 2026; detection only -- no structural verdict; echinocandins do not coordinate a metal, so the CYP51/heme-iron geometry does not transfer to FKS1. It sits in hot-spot HS2, at the R1354 anchor — a novel substitution at residue 1354, where S is a known echinocandin-resistance change.

How this works · methods & provenance

Source. Candida auris (Candidozyma auris) whole-genome deposits from NCBI Pathogen Detection, over a trailing 180-day window. The most recent batch re-called 3375 isolates over release PDG000000067.710. Public assemblies carry no echinocandin call (NCBI runs no AMR pipeline), so an out-of-band FKS1 re-call batch (cadence set by the ~97-day median deposit lag) produces the calls this page overlays.

Certified caller. The FKS1 re-caller is graded on a frozen external benchmark (fks1_benchmark_100.tsv, PMC12323592) on every change, through the same scorer this verdict uses:

Verdict
PASS (98/98 isolates resolved)
Sensitivity
100%
Specificity
100%
Exact-token identity
100%

Detection only — the echinocandin arm reports whether a known resistance variant is present, not a structural fit verdict. Research use only: a triage aid, not a clinical claim. Priority tiers and every number come straight from openafr.alert; this page renders that verdict, it never invents one.
